Aim: To give students a comprehensive knowledge to the experimental methods used in the modern molecular microbiology and physiology. There is a focus on questions relating to gene regulation and regulation of macromolecular synthesis. |
Contents: The course contains exercises in molecular microbiology and physiology. In the molecular part DNA is isolated from bacteria aiming at constructing gene libraries, cloning of genes in expression vectors, etc. The methods which are extensively used in modern molecular microbiological work are presented. Characterization of DNA using restriction enzymes and gel electrophoresis, gene splicing, PCR techniques, DNA sequencing, expression of genes, etc. In the physiological part, experiments are performed, which demonstrate various aspects of gene regulation and regulation of macromolecular synthesis (chromosome replication, transcription, translation). The final part of the course consists of free exercises, where the students, based on the results obtained during the course, plan experiments to solve a small reseach topic using the techniques presented during the course. Molecular microbiological and physiological topics of the departmental research are presented in lectures. |
