27805 Fungal chemo-taxonomy
analytical methods and practical applications
Danish title: Svampe kemotaksonomi
Language:  English    ECTS-creditpoints:  5
Type:  course at phd level
Program:  Chemical Engineering
Class schedule:
First time June 2003
Scope and form:  The course runs over three weeks with 15 days (9-16) that are divided between lectures, group discussions, practical laboratory work, seminars and small projects.
Evaluation:  Oral presentation and approval of reports
The course will be evaluated based on a written report/poster session and an oral presentation of the results of a small project
Participant limitation:  Max. 6
Aim:  Classification and taxonomy are traditionally based on morphological and anatomical characters, which are mostly easily observed. These classical morphological characters are important, but are often subjected to ecological modifications resulting in a high degree of plasticity among individuals of the same species. A more subtle set of characters, the secondary metabolites, have during the last century become increasingly important, for the taxonomy of fungi, particularly the moulds and the lichenized fungi. The use of chemosyndromes and secondary metabolite profiles for taxonomic analysis or species identification depend on a number of analytical methods and knowledge of the biosynthetic pathways of the metabolites.
Contents:  The course gives an introduction to modern theory and analytical tools in fungal chemotaxonomy. Special emphasize is put on moulds and lichenized fungi. The following topics will be covered:

Theoretical
o Principles of classification, systematics, taxonomy and cladistics
o The concept of species
o Secondary metabolites and their biosynthesis
o Introduction to analytical screening methods
o Introduction to chemometrics for taxonomy

Practical

o Isolation and cultivation of fungi
o Extraction of metabolites
o Separation, identification and quantification of secondary compounds using TLC, HPTLC, HPLC and LC-MS
o Chemo metrics and image analysis

Report and presentation techniques
Remarks:  A major part of the laboratory work will be on a small project, which to some extent can be a part of the participant own phd project.

Prerequist: High-level knowledge in one or more of the course topics corresponding to a master's degree and basic knowledge of the other topics including: organic chemistry, natural product chemistry, chromatography, mass spectrometry, fungal biology and taxonomy, systematics

Place:The course venue will be at BioCentrum-DTU, Building 221 Søltofts Plads, DK-2800 Kgs. Lyngby and Botanical Institute, University of Copenhagen, O. Farimagsgade 2D, DK-1353 Copenhagen, Denmark
